You can password-protect your profile to prevent somebody else from using your profile unless they know the password, using an extension listed below. This is easy to bypass by running the application in safe mode to temporarily disable all extensions. Somebody could also read your stored messages using Notepad since the messages for each folder are stored in text files ("inbox." is your inbox folder). However, this should be sufficient to keep casual users from using your profile.
Thunderbird
For Thunderbird there are two different extensions you can use.
- ProfilePassword password protects the whole profile.
- DiddlyDo password protects either the whole profile or just selected folders. The extension author has left the user interface deliberately vague, but see this forum thread for a few usage hints.

Firefox
To password-protect your Firefox profile, you can use the ProfilePassword extension.
